Abstract

Many plants and food supplements have been used for their properties against cancer for centuries. There have been many recent reports that natural compounds extracted from plants that can be used against cancer. The purpose of this research was to compare the extracts of five plant extracts against colon and breast cancer cells. Ashwagandha, Pomella (Pomegranate extract), Sesamin, Turmeric, and Cinnamon were selected because they were commonly used as natural foods. Ashwagandha is known as Indian ginseng. This Ayurvedic herb offers multiple therapeutic properties by helping with inflammation, stress, fatigue, and anxiety. Pomella is an extract of primary natural antioxidants present in pomegranate fruit. Sesamin is a lignan isolated from the bark of Fagara plants and from sesame oil. In addition, Turmeric and Cinnamon have been also used as spice on various types of food, drinks, and snacks. Although these entities have numerous therapeutic properties, their antiproliferative activities have not been compared with each other. Therefore, Ashwagandha, Turmeric, Pomella, Sesamin, and Cinnamon extracts from Verdure Sciences were tested against colon cancer cells (HT-29) and breast cancer cells (MDA-MB-468) using an MTS assay. MTS assay has been widely used as a colorimetric method for accurate quantification of viable cells in proliferation process. Among five tested extracts, Ashwagandha showed high antiproliferative activities at all concentrations (0.05-0.25 mg/mL) and reduced the growth of colon and breast cancer cells by 74-87%. The antiproliferative activities in colon cancer cells were significantly higher than breast cancer cells. Turmeric also reduced the proliferation of colon cancer cells by 75% at a concentration of 0.20 mg/mL but was less effective than Ashwagandha that reduced the proliferation by approximately 86% at the same concentration. Pomella slightly reduced the growth of both cancer cells at concentrations of 0.15-0.25 mg/mL by 22-33%. Cinnamon did not show high antiproliferative activity when compared with other extracts. Sesamin reduced breast cancer proliferation by 31.2% at 0.05 mg/mL while the cinnamon was ineffective. Thus, among five selected natural foods, Ashwagandha was the most effective extract with antiproliferative activities against colon and breast cancer cells while Cinnamon did not show any significant activity. The order of activity was Ashwagandha > Turmeric > Pomella > Sesamin > Cinnamon. More studies are required to study the antiproliferative activities of the most effective extracts, such as Ashwagandha and Turmeric in vivo to determine whether there is a correlation with the cell-based assays.
